Most prions (infectious proteins) are self‐propagating amyloids (filamentous protein multimers), and have been found in both mammals and fungal species. The prions [URE3] and [PSI+] of yeast are disease agents of Saccharomyces cerevisiae while [Het‐s] of Podospora anserina may serve a normal cellular function. The parallel in‐register beta‐sheet structure shown by prion amyloids makes possible a templating action at the end of filaments which explains the faithful transmission of variant differences in these molecules. We dispute Penn et al.'s claim of the sharp functional discontinuity between humans and nonhumans with evidence in bottlenose dolphins (Tursiops truncatus) of higher-order generalizations: spontaneous integration of previously learned rules and concepts in response to novel stimuli. We propose that species-general explanations that are in approach are more plausible than Penn et al.'s innatist approach of a genetically prespecified supermodule.

The nematode C. elegans exhibits a variety of reponses to touch. When specific sets of mechanosensory neurons are killed with a laser, specific touch responses are abolished. Many mutations that result in defective mechanosensation have been identified. Some of the mutations define genes that specify the fate of a set of mechanoreceptors called the touch cells, which mediate response to light touch to the body of the worm. Genes specifying touch cell fate appear to regulate genes that encode touch‐cell differentiation (...) proteins, including apparent subunits of a touch‐cell‐specific ion channel, rare mutant forms of which lead to swelling and lysis of the touch cells. Molecular attachments of the ion channel, both to extracellular matrix components and, intracellularly, to a special large‐diameter microtubule, may be required for mechanical gating of the channel. A mechanoreceptor‐interneuron‐motorneuron reflex circuit for response to light touch has been proposed. (shrink)

The US neutrino community gathered at the Workshop on the Intermediate Neutrino Program at Brookhaven National Laboratory February 4-6, 2015 to explore opportunities in neutrino physics over the next five to ten years. Scientists from particle, astroparticle and nuclear physics participated in the workshop. The workshop examined promising opportunities for neutrino physics in the intermediate term, including possible new small to mid-scale experiments, US contributions to large experiments, upgrades to existing experiments, R&D plans and theory. The workshop was organized into (...) two sets of parallel working group sessions, divided by physics topics and technology. Physics working groups covered topics on Sterile Neutrinos, Neutrino Mixing, Neutrino Interactions, Neutrino Properties and Astrophysical Neutrinos. Technology sessions were organized into Theory, Short-Baseline Accelerator Neutrinos, Reactor Neutrinos, Detector R&D and Source, Cyclotron and Meson Decay at Rest sessions.This report summarizes discussion and conclusions from the workshop. Cyclin-dependent kinase 5 has been implicated in Alzheimer's disease pathogenesis. Here, we demonstrate that overexpression of p25, an activator of cdk5, led to increased levels of BACE1 mRNA and protein in vitro and in vivo. A p25/cdk5 responsive region containing multiple sites for signal transducer and activator of transcription was identified in the BACE1 promoter. STAT3 interacts with the BACE1 promoter, and p25-overexpressing mice had elevated levels of pSTAT3 and BACE1, whereas cdk5-deficient mice had reduced levels. Furthermore, mice with a (...) targeted mutation in the STAT3 cdk5 responsive site had lower levels of BACE1. Increased BACE levels in p25 overexpressing mice correlated with enhanced amyloidogenic processing that could be reversed by a cdk5 inhibitor. These data demonstrate a pathway by which p25/cdk5 increases the amyloidogenic processing of APP through STAT3-mediated transcriptional control of BACE1 that could have implications for AD pathogenesis. Our starting point is a somewhat obscure incident which has lately attracted some attention. The year is 429 B.C., and the place is Athens in the third year of the Peloponnesian war. The plague, which had broken out only a year before, was still claiming its victims. Yet military operations were in full swing, and the general Phormio operating in the Corinthian gulf against a Peloponnesian fleet was able to score an impressive victory. The Lacedaemonians were deeply dissatisfied. This was (...) the first sea-fight they had been engaged in, and they found it hard to believe that their fleet was so much inferior to that of the Athenians. They dispatched three advisers to Knemos, the admiral in charge, instructing them to make better preparations for another sea-fight. Additional ships were solicited from the allies, and those already at hand were prepared for battle. It is at this point that the incident in question occurred. Not to prejudge the issue, I quote the text in full leaving the controversial phrases untranslated: 4. Pp. xii + 330.Modern scholarship on the Tao Te Ching has tended to focus on questions of authorship and the intended meaning of the text, often working from both the unquestioned assumption that matters of origination are of primary historical importance and the quasi-theological (...) bias that the original textual meaning is somehow the "right" or authoritative one. But the role of the Tao Te Ching in Chinese history and culture, as well as that of its purported author, is a much more complex affair. The text has been the subject of more than seven hundred commentaries and has been appropriated by vast networks of scholars, government officials, self-conscious religious communities, and laypeople, while Lao Tzu himself has undergone a fascinating history of deification and aesthetic representation. With Lao-tzu and the Tao-te-ching, editors Livia Kohn and Michael LaFargue take steps toward filling in some of the complicated (and much overlooked) post-canonical history by assembling essays on subjects ranging from the ritual uses of the text to modern hermeneutic experiments. The volume includes nine new essays, as well as three older ones reprinted or translated from earlier sources.The book is organized in four sections—"Ancient Myths," "Chinese Interpretations," "Modern Readings," and "Critical Methods"—although there is noticeable [End Page 625] overlap among the parts. The twelve essays are of varying quality and of varying use for different readers. Some of the material is highly technical and presupposes a sinological audience, while other material rehearses data that should already be familiar to scholars of ancient Chinese philosophy but may be tremendously useful to those who are interested in the text but lacking the scholarly background. One case of the latter is Julia Hardy's "Influential Western Interpretations of the Tao-te-ching," which does not especially break new ground but is a wonderful historical synopsis of Western scholarship on the text, as it meticulously identifies the key interpretive voices and sorts out their positions. Hardy gives only brief attention to the earliest eighteenth- and nineteenth-century scholarship—omitting, for example, any mention of Victor von Strauss' 1870 German rendition—which might actually have been most edifying for specialists. LaFargue and Julian Pas' "On Translating the Tao-te-ching" is essentially a primer on hermeneutic problems relevant to the text that serves as an intelligent corrective to the uncoordinated body of Taoist materials available to novice readers.What may be most clear from several of the essays taken in tandem is that the popular Western "mystical" reading of the text not only is one of many historical interpretations but is also very much heir to Wang Pi's intellectual lineage. In "A Tale of Two Commentaries: Ho-shang-kung and Wang Pi on the Lao-tzu," Alan K. L. Chan notes how such standard tenets as the spiritualized metaphysical understanding of Tao, the relationship between nonbeing and creativity, and the identification of naturalness and nonaction with spontaneity originate with Wang Pi but are nowhere to be found in the earlier Ho-shang-kung reading. The latter employs typical Han dynasty correlative cosmological interpretive methods, referring to naturalness as the preservation of energies in the physical body and to "the One" not as an abstract metaphysical oneness but as "the original substance of life itself, energy in its most pure and potent form" (p. 93). According to Isabel Robinet's "Later Commen-taries: Textual Polysemy and Syncretistic Interpretations," the panoply of text interpretations falls into roughly three categories—"technical or specialist readings," "philosophical exegeses," and "inner alchemy interpretations," which she then proceeds to divide further into "philological" and "ideological" readings. This is all further complicated when one considers the Tao Te Ching not as a strictly philosophical text but as a collection of polemical aphorisms, as LaFargue does in "Recovering the Tao-te-ching's Original Meaning: Some Remarks on Historical Hermeneutics," or as a ritual, liturgical or meditative aid, as Kohn does in "The Tao-te-ching in Ritual."Lao-tzu and the... Gandhi, Martin Luther King Jr., and Gautama the Buddha in the Twenty-First Century. By A. L. Herman. Albany: State University of New York Press, 1998. xi + 245 (...) pp. At first glance, this title might seem a bit over the edge: how could an author intelligently juxtapose such diverse thinkers as an ecological scientist, two advocates of human rights, and an ancient religious founder? For the most part, the author succeeds in demonstrating that the twenty-first century requires a definition of community that includes environmental, social, and spiritual concerns. In a format particularly [End Page 265] helpful for undergraduate teaching, he carefully rehearses at the end of each section three central questions: how does this thinker contribute to our understanding of community? Does his approach lead to the diminishment of violence? Does it facilitate peace?For the first quarter of the book, Herman explores various aspects of community. Drawing from Erich Fromm, Socrates, and Aldous Huxley, he demonstrates that out of enlightened self-interest, all people seek peace for themselves. He is careful to point out, however, that the Socratic virtues of wisdom, courage, self-control, and justice, which arise from within, are far superior to the uniformity of the World State as described in Huxley's Brave New World, which leaves nothing uncontrolled by the external authorities. Both lead to nonviolence and peace, but the World State can fall prey to what Fromm terms cultural insanity.Supplementing his discussion of Aldo Leopold with references to the work of Rachel Carson and J. Baird Callicott, Herman emphasizes that ideas of community peace must be informed by holistic structures of nature, particularly as articulated in Leopold's Sand County Almanac. However, he also warns that if the welfare of the biotic community takes precedence over all other concerns, then society could fall into a state of ecofascism.Herman's chapter on Mahatma Gandhi ably summarizes the life and works of this great political reformer, with particular focus on his formative years in South Africa. Influenced by John Ruskin and Leo Tolstoy, Gandhi developed the concept of the "Ashramic community," a means of collective, intentional living designed to effect social change. After describing the Phoenix Settlement (1904, Johannesburg), the Sabarmati Ashram (1915, Ahmedabad), and Sevagram Ashram (1933, Wardha), Herman presents a fifteen-page play script based on the Asian travel diaries of Professor George P. Conger, chair of philosophy at the University of Minnesota from 1937-1953. Conger spent two months at Gandhi's ashram in Wardha, India, in 1933, and in the play called "Salt," his students take up issues including the ill treatment of Jews in Germany, the definition of nonviolence, and emphasized, as does Gandhi, the necessity for self-transformation to achieve personal and community peace.Martin Luther King Jr. inspired by the example of Mahatma Gandhi, employed nonviolent techniques to bring about the many successes of the American Civil Rights Movement. Herman provides a brief biography of Martin Luther King Jr., quotes several of his pivotal sermons, and emphasizes the notion of beloved community as critical to King's thinking. He writes that "in King's interpretation of agape, it is your being loved that gives you value; and it is your selflessly loving others that gives them value and makes the beloved community possible" (p. 138). According to King, fear and racial discrimination prevent one from entering into the beloved community. By changing laws, and, more importantly, changing the hearts of individual people, a more peaceful community can be built.The least successful chapter of this book discusses Gautama Buddha and the "karmic community." Rather than focusing directly on Buddhist theories of Karma, Herman engages in a long discussion of the Bhagavad Gita, an important Hindu text, as a foil to explain Buddhist theories... (shrink)

The ability of the mind to conceptualize what is not present is essential. It allows us to reason counterfactually about what might have happened had events unfolded differently or had another course of action been taken. It allows us to think about what might happen – to perform 'Gedankenexperimente' (thought experiments) – before we act. However, the cognitive and neural mechanisms mediating this ability are poorly understood. We suggest that the frontopolar cortex (FPC) keeps track of and evaluates alternative choices (...) (what we might have done), whereas the anterior lateral prefrontal cortex (alPFC) compares simulations of possible future scenarios (what we might do) and evaluates their reward values.
